β-catenin is a multifunctional protein, its main function is to mediate cell adhesion and participate in the regulation of gene expression, and its expression is closely regulated by APC and other related proteins; recent studies have shown that due to APC and its related genes The mutation that leads to abnormal accumulation of β-catenin is closely related to the occurrence of colon cancer. Therefore, looking for a new APC-independent β-catenin expression regulation pathway is extremely important for the treatment of colon cancer. The study found that the orphan nuclear receptor Nur77 can negatively regulate the expression of β-catenin through its non-genotypic function, thereby inhibiting the growth of tumor cells. At the same time, the study also found two cardiac glycoside compounds that can negatively regulate the expression of β-catenin by inducing the expression of Nur77: H-9 and ATE-i2-b4. The study proves that Nur77 can be used as a potential target for colon cancer treatment at the molecular, cellular and animal levels, and H-9 and ATE-i2-b4 may become potential drugs for the treatment of colon cancer.
